DATA ANALYSIS, DATA PRESENTATION AND INTERPRETATION
BACKGROUND
A new experimental autoimmune encephalomyelitis model of multiple sclerosis has been developed in Mus
spretus and animals have been treated with rolipram (a phosphodiesterase-4 inhibitor) to test the hypothesis
that rolipram will inhibit the development of disease.
METHODS
8-10 week female adult Mus spretus Were purchased from Envigo, Bicester, UK and animals were housed
and used in accordance with the Animals (Scientific procedures) Act 1986 incorporating ethical review.
Animals were injected with 200pg myelin oligodendrocyte glycoprotein peptide residues 35-55 emulsified in
Freunds complete adjuvant on day O and 7. Animals were monitored daily from day 10-25. Neurological
scores were assessed in a blinded fashion and were scored normal; 1 limp tail; 22 impaired righting
reflex (when you turn animal on its back it cannot right itself); 2 Hindlimb paresis; 4 Hindlimb paralysis,
5 Forelimb paralysis. Rolipram was purchased from Sigma, Poole Dorset and dissolved in 5% dimethyl
sulphoxide in phosphate buffered saline (This vehicle is known not to influence the disease course) and
20mg/kg was injected intraperitoneally daily from day 11 onwards. Animals (n:10/group. based on sample
size calculations from previous work) were randomised to treatment or vehicle.
RESULTS

EAE was induced on day O. Animals were treated with either vehicle (V) or 20mg/kg rolipram (R)
